Koesobivusantigeenid osalevad keerulises immuunkompleksis, mille abil suudab organism eristada \u201eoma\u201c ja \u201ev\u00f5\u00f5rast\u201c. \u201eV\u00f5\u00f5rast\u201c valku kandvad rakud h\u00e4vitatakse T-l\u00fcmfots\u00fc\u00fctide poolt. Inimesel nimetatakse MHC valke HLA antigeenideks (human-leucocyte-associated antigens), kuna esmalt avastati nad leukots\u00fc\u00fctidel.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">HLA geenikompleks sisaldab endas mitmeid lookusi: A, B ja C on peamised klass I geenilookused ning DR, DQ, DP on peamised klass II geenilookused. Klass I valgud on esindatud enamikul rakkudel, klass II valgud ainult antigeene esitlevatel rakkudel. HLA geenid paiknevad 6. kromosoomis ning need on k\u00f5ige pol\u00fcmorfsemad geenid inimese genoomis: 2025. aasta oktoobri seisuga on teada 42193 erinevat alleeli ehk geenivarianti. HLA alleelid p\u00e4randuvad: \u00fcks alleelide komplekt saadakse emalt ja teine isalt. \u00d5dedel-vendadel on identsete HLA alleelikomplektide esinemist\u00f5en\u00e4osus 25%.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">HLA alleelide m\u00e4\u00e4ramisv\u00f5imalused \u00dchendlaboris:<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">madallahutusega meetodid:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>SSO \u2013 j\u00e4rjestusspetsiifiline pol\u00fcmeraasi ahelreaktsioon ja j\u00e4rjestusspetsiifiliste oligonukleotiidproovidega h\u00fcbridisatsioon;<\/li>\n\n\n\n<li>RT PCR \u2013 reaalaja pol\u00fcmeraasi ahelreaktsioon;<\/li>\n<\/ul>\n\n\n\n<p class=\"wp-block-paragraph\"><em>k\u00f5rglahutusega<\/em> meetodid:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>NGS \u2013 j\u00e4rgmise p\u00f5lvkonna sekveneerimine.<\/li>\n<\/ul>\n\n\n\n<p class=\"wp-block-paragraph\">Madallahutusega t\u00fcpiseerimise korral tehakse kindlaks alleeligrupp ehk kaks kohta alleeli t\u00e4hises (nt HLA-A*02); k\u00f5rglahutusega t\u00fcpiseerimise korral m\u00e4\u00e4ratakse alleeli t\u00e4hises 4 kuni 6 numbrikohta (nt HLA-A*02:01:01).<\/p>\n\n\n\n<p class=\"has-large-font-size wp-block-paragraph\"><strong>HLA alleelide m\u00e4\u00e4ramine elundite siirdamiseks<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Elundite siirdamise eelselt on vajalik m\u00e4\u00e4rata nii
